De ascendencia siciliana, griega, árabe, francesa, irlandesa y alemana, fue el mayor de cuatro hijos. A causa del asma que Frank sufría, su familia se mudó en enero de 1951 a Monterey, California, unos 150 kilómetros al sur de San Francisco. Antes de volver a California (San Diego) a principios de los años 50, vivieron un tiempo en Pomona y en El Cajón. En 1955 la familia se reestableció en Lancaster, pequeña ciudad en el valle Antelope, en el desierto Mojave, unos kilómetros al norte de Los Ángeles. A los 15 años, Frank había pasado por seis escuelas diferentes, lo que quizá contribuyó al sentido de alienación que lo embargaría en su edad adulta.Su padre, que había nacido en Sicilia, era químico y trabajaba cerca de la base Edwards de la Fuerza Aérea estadounidense, lugar dedicado en aquella época a la investigación en el campo de las armas químicas. Dada esa proximidad, tenía en su casa máscaras de gas, en caso de que hubiera algún accidente y es seguro que esto afectó enormemente al joven Frank: en su obra se encuentran varias referencias a gérmenes, guerra bacteriológica y otros aspectos de la industria militar secreta.
La situación de Lancaster permitió a Frank acceder a la estimulante influencia de los sonidos (musicales y publicitarios) de las radios de Los Ángeles y zonas aledañas, y sus padres le compraron un tocadiscos, discos, un televisor e instrumentos musicales. La televisión fue asimismo otra gran influencia: en casi todas sus obras hay referencias a ella, a sus programas, sus anuncios publicitarios y sus jingles.
Su obra cubrió una larga variedad de estilos musicales (principalmente, avant-garde, rock
psicodélico, jazz fusión, y clásico contemporáneo) y se destacó en su mezcla de academia, ópera rock, absurdo, humor escatológico y satírico.Creó y lideró una gran banda musical The Mothers Of Invention, posteriormente llamados solo "Mothers", por la que pasaron los mejores exponentes del rock y el jazz de su generación, un exquisito grupo de músicos entre los que destacan Jean Luc Ponty, George Duke, Warren Cuccurullo y otros.
